Your credit score is based on what is in your credit file currently. Once an item is no longer on your credit report it is like it never happened.
Depending on your situation and what was removed your score may go up dramatically, it may go up a little or not at all. But it will change as soon as the information is removed.
Paying it, alone, will not affect your credit score. The good news is this: Once an item is paid off with a collection agency, it is relatively easy to get removed. Check which Credit Reporting Agencies that the collection is reporting to and dispute it with them. The reason for this is that once the collection agency has their money, that's all they care about. If you dispute it they would have to take the time to verify the debt and send a response back to the CRA. Collection agencies aren't exactly at the top of the moral pyramid when it comes to this stuff. They will deal from the bottom of the deck to collect, but when it comes time to spend money to research the debt and verify it, they usually don't do it.
