I don't know for sure. My answer is I don't think it really makes a difference. You could call the human resource dept. at another company and ask them. I think they will tell you it doesn't matter. Probably more important is a follow up letter from you, (not email). After you apply, send a letter stating you have applied and your interest in the job and how much you hope to interview and so on. Make it a short letter addressed to the correct person. If you don't have a name, call the company and ask to whom your letter should be directed.

Normally, the ad for the job will tell you how to apply. If you can apply in person, it shows the employer that you are willing to take a few extra steps in making sure they know who you are.

Some places only let you apply online, but either way you do always do follow ups with the company, a simple phone saying "i am just calling to do a follow up on my application" is the best thing to do, if you had the chose to go into the store and apply thats the best b/c then you can hand the application into a manager and they will know more about whether or not you have a good shot at the job and they can set you up with an interview.

It 100% depends on the instructions. Follow the instructions. When my company was looking for a new CEO, I had people coming right into my office trying to pitch themselves, and some were very impressive, but regardless it became rather annoying, and I didn't give any of them any extra consideration. The woman who we did end up choosing simply submitted her resume and cover letter, just as the advertisement had requested. So do whatever the ad says and follow instructions. Present a very poignant, subtly persuasive and concise to-the-point cover letter and a resume that shows you're the right person for the job (assuming of course that you are the right person), and you will get past that hurdle and onto the interviews!
I have applied many times online, and about 85% of the jobs that i've applied have called me for an interview. Just make sure you apply to those secure websites, like monster.com and stuff like that and they should call you. But of course, if you have the possibility to apply in person, then go for it!
At my job I like the people that show iniative and come in in person I think that gives them a feel for the business also. You may have more success also if you like the environment and can feel it out, rather than over the internet you don't know what to expect.
